Design and Development of Release Modulated Anti-inflammatory Topical Gel

ABSTRACT
Celecoxib is a nonsteroidal anti-inflammatory drug (NSAID) belonging to BCS II, used in the treatment of rheumatism and osteoarthritis. In this research work an attempt was made to develop a release modulated anti-inflammatory topical gel loaded with Celecoxib microbeads. Solubility of poorly soluble Celecoxib was enhanced by preparing inclusion complex with 2-Hydroxypropylβ-Cyclodextrin using kneading method. The best fit formulation (HF3) with acceptable solubility was incorporated in the microbead formulations prepared by inotropic gelation method. Drug loaded microbeads were incorporated in the gel prepared using varying concentrations of Carbopol 934P. The pH of prepared microbead loaded gel formulation was found to be within the acceptable range. The prepared formulations showed good spreadability and drug content which was found to be within pharmacopoeial limit. The results of in vitro drug release studies revealed that the formulation releases more than 90% of drug extended over a period of 12 hours.
