Effects of cold scarification and GA3 at 4ºC on seed Germination of Meizotropis pellita a wild species of Indian Himalayan Region

ABSTRACT
Meizotropis pellita is an endemic plant species of Uttarakhand. Meizotropis pellita seeds are broad and unorthodox in nature. Seeds of Patwa are Dormant even under natural environment and this phenomenon is described as Dormancy. Meizotropis pellita has low germination rate due to hardness and impermeability of the seed coat. To overcome seed dormancy different Stratification treatments is used. The sterilized seeds were kept at 4ºC and pre-soaked in GA3 (.05g/l) at 4ºC for different durations. Then treated seed were transferred to the conical flask containing MS media for germination. The results of the present study showed that Cold Scarification and GA3 at 40C were necessary to break dormancy and to enhance germination of Meizotropis pellita seeds at different time intervals. It was found that on Treatment Cold Scarification at 4⁰C a visible breakage of seed coat with in 24 hrs and on Treatment GA3 at 4⁰C for 24 hrs gave 100% germination. Thus, this present work will offer an efficient in vitro seeds germination of Meizotropis pellita for Conservation.
