Evaluation of Anti-gouty arthritis and Anti-hyperuricemic activities of Cucurbita maxima seeds extracts on Potassium oxonate and Monosodium urate crystals induced Gout in Sprague Dawley rats

ABSTRACT
The present study has been conducted to evaluate the Anti-hyperuricemic and Anti-gout activity of ethanolic extract of Cucurbita maxima seed extracts on potassium oxonate and Monosodium Urate crystals induced gout in Sprague Dawley rats. Sprague Dawley rats were divided into 5 groups of 6 each. In Hyperuricemic model, except control group all the groups were induced with hyperuricemia on the administration of Potassium oxonate (250mg/kg/i.p) for 21 days. Allopurinol was used as a standard drug(5mg/kg/p.o). EECM at low dose(200mg/kg/p.o) and high dose(400mg/kg/p.o) were administered orally for 21days. On 21st day, all the groups of rats were anaesthetized and sacrificed. The blood serumwas separated and the liver was isolated and homogenized. Serum urate level and xanthine oxidase level were estimated. The body weight was also recorded on day 1,7,14 and 21. In gout model, the study was conducted for 9 days. Colchicine 0.3mg/kg/p.o) was used as a standard drug. EECM at low dose(200mg/kg/p.o) and high dose(400 mg/kg/p.o) were administered for 9 days. After 1 hour,except control group all the groups were injected with Monosodium urate crystals (3 mg/kg/I-ARTI) on right knee on last 3 days. On 9th day right knee circumference in all the groups were measured at 0, 12, 24, and 48 h after the last dose of MSU crystal injection and the swelling index was calculated also the histopathological changes were analyzed in terms of diminished joint space, deformation of joint synovium, and inflammatory cells infilterates in the joints. The results of EECM treatment revealed that, elevated serum urate level and body weight in group IV and V were significantly decreased compared to hyperuricemic group and xanthine oxidaseinhibition were significantly increased in group IV and V compared to hyperuricemic group. Swelling index in group IV and V were decreased when compared to group III also the histopathological study showed the normal architecture, reduction in synovial tissue swelling and cellular infilteration. Overall, from the obtained results, it is determined that the ethanolic extract of Cucurbita maxima seeds exhibited a good Anti-hyperuricemic and antigouty arthritic effect.
