Gastroretentive Drug Delivery System

ABSTRACT The gastro-retentive drug delivery system (GRDDS) has recently become highly favored in the realm of oral drug delivery. This approach effectively retains the dosage form in the stomach for a prolonged duration and allows for a gradual release of the drug, addressing various issues linked to traditional oral delivery methods, such as low bioavailability. A range of innovative strategies, including magnetic field-assisted gastro-retention, plug-type swelling systems, muco-adhesion techniques, and floating systems with or without effervescence, are being utilized to develop GRDDS. In addition to in vitro characterization, the successful creation of GRDDS necessitates carefully designed in vivo studies to confirm improved gastro-retention and extended drug release. Gamma scintigraphy and MRI are well-established methods for assessing in vivo gastric residence time. Nonetheless, evaluating their overall in-vivo effectiveness continues to pose a significant challenge for this type of dosage form, particularly in small animals such as mice or rats. Despite a large number of promising in vitro findings, published in vivo studies involving beagle dogs, rabbits, and human participants are quite limited.
