Isolation and Characterization of Indole Acetic Acid (IAA) Producing Bacteria from Rhizospheric Soil and It’s Effect on Plant Growth

ABSTRACT

Indole acetic acid (IAA) production is a major property of plant growth promoting rhizobacteria  (PGPR) that stimulate and facilitate plant growth. The present work deals with isolation, characterization, optimization and identification of indole acetic acid producing bacteria from the rhizospheric soil. Out of Six Indole acetic acid producing isolates, two were selected as efficient producers. Optimization of indole acetic acid production was carried out at different cultural conditions of pH and temperature with varying media components such as carbon and nitrogen source concentration. The concentration of indole acetic acid was carried at different cultures and purification of IAA was done and purity was confirmed with Thin Layer Chromatography (TLC) and High performance liquid chromatography (HPLC). All the two isolates were identified by amplifying their 16S rRNA gene sequencing. Subsequently, effect of plant growth was tested by pot assay. Biofertilizer prepared by using IAA producing bacterial isolates mixed with Charcoal in 1:3 ratio. In conclusion of the study was IAA producing bacteria as efficient biofertlizer inoculants to enhance plant growth that which increase their growth of stem and leaves.
