Simultaneous Quantification of Five Markers from a Mangrove Avicennia Marina, by Uni-Dimensional Double Development HPTLC and Scanning Densitometry

ABSTRACT

Avicennia marina (Forsk.) Vierh. is a common mangrove traditionally reported to be used in curing a range of ailments such as rheumatism, small pox, ulcers, etc. Scientific data on the phytochemistry, and therapeutic potential of this plant is scarce. In the present research work, Uni-Dimensional Double Development HPTLC method has been developed for the simultaneous estimation of five potent biomarkers viz. ursolic acid, betulinic acid, betulin, betasitosterol and lupeol from leaves of Avicennia marina.

Chromatographic separation was achieved on 60F254 HPTLC plate with toluene: acetone as mobile phase after double development and derivatization with 10% methanolic sulphuric acid reagent. The method was validated as per ICH guidelines. The regional variation in the marker content was evaluated by assessing samples collected from ten geographically variant locations. The total combined content of these five markers was found to be highest in the sample collected from Airoli, Mumbai.

The band pattern established during the research work can also be used as a fingerprint and may assist in quality evaluation and standardization in absence of markers. This data can also be used to establish a cross-talk between chromatographically established phytochemistry and bioassays to provide an assurance of efficacy and consistency of plant extracts.
