Studies on Production, Optimization and Purification of Industrially Important Laccases and Their Application in Dye Degradation & Bio-Transformation

Laccases are the versatile enzymes which catalyze oxidation reactions coupled to four-electron reduction of molecular oxygen to water. They are multicopper enzymes capable of degrading lignin and are present abundantly in higher plants and white-rot fungi. However Bacterial laccases have several unique properties that are not characteristics of fungal laccases such as stability at high temperature and high pH. They decolorize the dyes and detoxify the industrial effluents and help in wastewater treatment. Laccase has been applied to nanobiotechnology which is an increasing research field and catalyzes electron transfer reactions without additional cofactors. Recently several immobilization techniques have been developed such as micro patterning, self-assembled monolayer, and layer by layer technique which immobilize laccase and preserve their enzymatic activity. Hence laccase is receiving much attention of researchers around the globe. To meet the current demand of the industry’s requirement it is necessary to increase the yield by optimizing various parameters which can produce higher titers of this enzyme. In the present study crude enzyme from Pseudomonas alcaliphila was produced under Submerged conditionsby addition of Copper Sulphate for enhanced laccase production and to evaluate its potential in dye degradation.
