Understanding Migraine: Pathophysiological Mechanisms and Modern Therapeutics

ABSTRACT Migraine is a frequent, severely disabling neurological illness with a complicated pathophysiology that involves a number of central and peripheral nervous system regions and networks. Primary recurring headaches lasting 4 to 72 hours with at least two of the following pain characteristics—unilateral, pulsing, moderate or severe intensity, or worsened by ordinary physical activity—are the hallmark of migraine, a common and frequently incapacitating neurologic illness. The purpose of this study was to evaluate general understanding of migraine pathophysiology by examining the structure and function of the primary disease-causing areas, with an emphasis on how these contribute to the wide range of symptoms that define attacks and the illness as a whole,Therapeutic treatments that have evolved for the treatment of migraine include compounds targeting calcitonin gene-related peptide (gepants and monoclonal antibodies), serotonin 5-HT1F receptor agonists (ditans), and non-invasive neuromodulation,as well as giving a synopsis of recent research supporting traditional migraine therapies.
