ABSTRACT Heterocyclic structures have emerged as pivotal elements in the development of anticancer agents due to their diverse chemical reactivities and biological activities. This review explores the role of heterocyclic compounds in anticancer drug discovery, emphasizing their mechanisms of action and clinical implications. It examines how various heterocycles, including pyridines, pyrimidines, and imidazoles, contribute to therapeutic efficacy through interactions with molecular targets involved in cancer cell proliferation and survival. The review highlights recent advancements in heterocyclic drug design, showcasing promising candidates that have progressed to clinical trials. It also discusses the challenges and future directions in optimizing heterocyclic compounds for better selectivity, reduced toxicity, and enhanced efficacy. This comprehensive analysis underscores the importance of heterocycles in shaping the future of cancer therapy.
