A Statistical Analysis of TP53 Mutation Impact on Survival in Lung Adenocarcinoma

ABSTRACT The prognostic significance of TP53 gene mutations in lung adenocarcinoma is assessed in this study using survival analysis. We estimated survival probability distributions for patient cohorts with wild-type versus mutated TP53 using the Kaplan-Meier method, and we used the Log-Rank test to determine the statistical significance of observed survival differences. Our analysis showed that patients with TP53 mutations had a significantly lower overall survival (Log-Rank test: χ² = 12.7, p = 0.0004), with a median survival of 3.2 years compared to 5.8 years for patients with wild-type TP53. The calculated hazard ratio of 1.65 (95% CI: 1.25 2.18) indicates a 65% increased risk of mortality associated with TP53 mutation. These findings highlight the critical importance of statistical methods in validating molecular biomarkers for cancer prognosis.
