Cytochrome P450: A Comprehensive Review of Its Role in Drug Metabolism, Interactions, And Disease Implications"

Author Name : Sakshi Gaikwad, Ms Pooja Karvande, Dr. Gajanan Sanap

ABSTRACT Drug-Drug interaction have come an important issue in health care. It's now realized that numerous medicinemedicine relations can be explained by differences in the metabolic enzymes that are present in the liver and other extrahepatic apkins. Numerous of the major pharmacokinetic relations between medicines are due to hepatic cytochrome P450 enzymes being affected by former administration of other medicines. After coadministration, some medicines act as potent enzyme corrupters, whereas others are impediments. still, reports of enzyme inhibition are veritably much more common. Understanding these mechanisms of enzyme inhibition or induction is extremely important in order to give applicable multiple- medicine curatives. In future, it may help to identify individualities at topmost threat of medicine relations and adverse events. The cytochromesP450 (P450) are a large group of native and inducible haem-containing enzymes, which have a central part in the oxidative metabolism of a different range of xenobiotic. Several P450 genes are polymorphic and are associated with the increased threat of cancer development in specific apkins. Individual P450s, especially CYP1B1, are over expressed in different types of tumors. The increased expression of P450s in tumors is largely significant and is important for understanding tumour development and progression. The tumourspecific expression of P450 provides the base for the development of new individual and remedial strategies. Besides the liver and other supplemental organs, P450 isoforms are expressed in glial cells and neurons of the brain. As millions of cases with HIV/ AIDS are put on treatment with the largely active antiretroviral remedy( HAART), medicine relations have come a major concern for healthcare providers. The use of HAART as a combination of 3-4 medicines creates implicit for antiretroviral( ARV) medicine relations, and this is complicated by the addition of other medicines for treatment of other affections similar as comorbid habitual conditions and/ or opportunistic infections.
