Development and validation of HPLC method for simultaneous determination of catechol, caffeic acid and anthraquinone in leaf extract of Xanthium strumarium and its Formulation.

ABSTRACT A sensitive, simple, and accurate high-performance liquid chromatographic method has been developed for simultaneous determination of Catechol, Caffeic acid and Anthraquinone in the leaf powder of Xanthium strumarium Linn and thereby provided a quality control tool to an anti-inflammatory formulation powder available in local market which contains leaf extract of Xanthium strumarium. The optimal analytical conditions are investigated to obtain the best resolution and the highest UV sensitivity for the quantitative detection of above mentioned components. The optimized conditions (Mobile phase A: 0.01 M phosphate buffer; pH 3 adjusted with ortho phosphoric acid And Mobile phase B : Acetonitrile(100%) , gradient elution system on a phenyl reversed-phase column with a flow rate of 1 mL/min and UV absorbance at 210 nm) allowed a specific and repeatable separation of the studied analytes.Parameters for the validation included linearity, precision, accuracy, recovery, Limit of Detection and Limit of Quantitation (LOD and LOQ).
