ABSTRACT This research article presents the development and validation of a UV spectrophotometric method for the determination of allopurinol in both bulk drug and pharmaceutical dosage forms. Allopurinol, a xanthine oxidase inhibitor, is widely used for treating gout and hyperuricemia(1,2). The study focused on developing a simple, cost-effective, and sensitive UV spectrophotometric method to ensure accurate and reliable quantification of allopurinol. Methanol was selected as the solvent, and the standard solutions were scanned in the range of 200-400 nm, with allopurinol showing maximum absorbance at 249 nm(3). Calibration curves were prepared with concentrations ranging from 0.4 to 2.0 µg/ml, demonstrating excellent linearity (R² = 0.999). The method was validated as per ICH guidelines, with accuracy confirmed through recovery studies at different concentration levels (98-102% recovery), and precision established by repeatability tests and intra-day and inter-day precision studies. The method was successfully applied to the analysis of marketed formulations, specifically Zyloric 100 mg tablets, showing the percentage of label claims within the range of 98-101%. The validated method provides a reliable tool for routine quality control and regulatory compliance, ensuring the therapeutic efficacy and safety of allopurinol-containing products.
