Emerging Anti-Angiogenic Strategies: An Updated Review

ABSTRACT Angiogenesis, the formation of new blood vessels from pre-existing vasculature, is essential in both physiological processes and pathological conditions such as cancer. While anti-angiogenic therapies—particularly those targeting the VEGF/VEGFR axis—have provided clinical benefit, their efficacy remains limited by resistance, toxicity, and a lack of reliable predictive biomarkers. Tumors can evade VEGF blockade through alternative signaling pathways, vessel co-option, or vasculogenic mimicry, necessitating the development of novel therapeutic approaches.
