Evaluation of Antidiabetic and Antioxidant Activities of Robinia pseudoacacia Leaves Using DPPH, Yeast Glucose Uptake Assay and GC–MS Analysis

ABSTRACT Diabetes mellitus is a chronic metabolic disorder characterized by persistent hyperglycemia and increased oxidative stress, contributing to the development of severe diabetic complications. The present study aimed to evaluate the antidiabetic and antioxidant activities of Robinia pseudoacacia leaves using in vitro experimental models. Ethanolic extract of R. pseudoacacia leaves was prepared by maceration and subjected to preliminary phytochemical screening, GC–MS analysis, DPPH free radical scavenging assay, and yeast glucose uptake assay. Phytochemical screening revealed the presence of flavonoids, tannins, phenolics, saponins, alkaloids, and glycosides. GC–MS analysis identified several bioactive compounds with reported antioxidant and hypoglycemic potential. The extract exhibited significant dose-dependent antioxidant activity and enhanced glucose uptake in yeast cells, indicating antidiabetic activity. The findings suggest that R. pseudoacacia leaves possess promising antioxidant and antidiabetic properties and may serve as a potential natural therapeutic agent for diabetes management.
