Formulation and Characterization of Fast-Dissolving Tablets of Promethazine Hydrochloride

ABSTRACT Promethazine HCl fast dissolving tablets (FDTs) are made by combining various both synthetic and natural super disintegrants. Aegle marmelos (bael), Plantago ovata (ispaghula), and dried banana powder mucilage were extracted and evaluated by a variety of physicochemical analyses. The following parameters were measured in that order: pH, stability, loss of drying, moisture absorption, melting point, and swelling index. Promethazine hydrochloride FDTs were produced using the direct compression technique with varying concentrations (5–15%) of Aegle marmelos (bael) and Plantago ovata (ispaghula) mucilage, as well as dehydrated banana powder, acting as natural superdisintegrants. These were then compared to sodium starch glycolate, a well-known synthetic superdisintegrants. Post-compression parameters such as thickness, hardness, friability, weight variation, wetting time, active ingredient content (%), in-vitro disintegration, in-vitro dispersion, and in-vitro dissolution have been evaluated for the manufactured tablets. The developed tablets underwent several types of physical testing, including thickness, hardness, friability, weight variation, and compliance with limitations. The formulation F3, which contains 15% of Plantago Ovata, has the lowest disintegration time (18.00 seconds) and dispersion time (29.00 minutes), resulting in a greater drug release rate of 95.43% in 12 minutes. It is therefore regarded as an optimal formulation. The results of this investigation showed that, as a natural superdisintegrant, Plantago ovata exhibited superior disintegration qualities than sodium starch glycolate, a synthetic superdisintegrant that is most frequently utilized in the formulation of FDTs. According to research, Plantago ovata functions well as a super disintegration agent.
