ABSTRACT Transdermal drug delivery systems (TDDS) have emerged as a promising and widely accepted approach for systemic drug administration through intact skin. This method offers several advantages over conventional oral and parenteral routes, including improved patient compliance, sustained drug release, and avoidance of first-pass metabolism. Topical administration via the skin enables both localized and systemic drug effects, making it a desirable alternative for drugs with limited oral bioavailability or gastrointestinal side effects. The aim of present study was to formulate and evaluate Matrix type transdermal drug delivery system (TDDS) of Donepezil, used in the management of Alzheimer’s disease was prepared by the solvent evaporation technique, aiming to achieve controlled and sustained drug release through the skin. Several batches were prepared by using combination of HPMC-E50, Eudragit-RS100 and PVP K90 different ratios. PEG-400 was used as plasticizer and DMSO was incorporated as a permeation enhancer.
