Improving Paracetamol Solubility Using Solid Dispersion Technique

ABSTRACT: Paracetamol is popularly employed drug in various types of pain. Paracetamol is sparingly soluble in water, which hampers the dissolution rate of this drug in the biological fluid. This results in poor bioavailability of Paracetamol ones it is orally administrated. In pharmacology, Bioavailability of the drug means the portion of drug that reaches the systemic circulation.When the drug is administrated intravenously, its bioavailability is 100%.However, that is not the situation when the drug is taken orally, especially in the case of drugs like paracetamol which is poorly water-soluble drug. Many techniques and methods have been implemented to enhance the bioavailability of the sparingly soluble drugs.This study incorporates the use of polyethylene glycol (PEG 8000) to enhance bioavailability of paracetamol. Paracetamol and polyethylene glycol (PEG 8000) were used in 1:1, 1:2, 1: 3 and 1: 4 ratios to study the effect of polyethylene glycol (PEG 8000) in dissolution profile of drug. The SOLID DISPERSION TECHNIQUE was used in this study. Paracetamol and PEG 8000 was mixed by using the kneading method to study the dissolution profile of drug. The study showed that by using the kneading method, the rate of dissolution of paracetamol was increased in PCM and PEG 8000 with the proportion of (1:4).
