Niacinamide: Multifunctional Molecule in Cosmetics and Therapeutics

ABSTRACT Niacinamide, a water-soluble derivative of vitamin B3has gained significant attention for its versatile cosmetic and therapeutic properties. As a key molecule involved in the biosynthesis ofnicotinamide adenine dinucleotide, it plays an essential role in maintaining cellular energy, DNA repair and the modulation of oxidative stress. Its potent antiinflammatory, antioxidant, and barrier-strengthening effects have led to widespread use in dermatology and cosmetic formulations. In cosmetics, niacinamide is widely acclaimed for its ability to improve skin hydration, reduce appearance of fine lines, wrinkles and brighten skin tone by inhibiting the transfer of melanosomes to keratinocytes. It is also effective in reducing hyperpigmentation and improving overall skin texture. Niacinamide’s ability to regulate sebum production and reduce inflammation makes it a popular ingredient in products targeting rosacea and acne. Furthermore, its barrier-repairing properties enhance the skin's resistance to external irritants, promoting improved resilience against environmental damage Therapeutically, niacinamide has shown promise in treating several dermatological conditions including atopic dermatitis, eczema and melasma. Its anti-inflammatory properties help manage inflammatory skin disorders, while its antioxidant activity protects skin cells from oxidative damage caused by UV radiation and pollution. This review explores clinical studies, dosage considerations, formulation challenges, and future prospects of niacinamide as a multi-functional ingredient in both cosmetic and therapeutic settings.
