Research on the Synthesis of Dyclonine Hydrochloride

ABSTRACT A new synthetic route to dyclonine hydrochloride is presented in this study. First, n-butoxybenzene was obtained via the reaction of phenol with bromobutane. Subsequent Friedel-Crafts acylation with 3-chloropropionyl chloride yielded 1-(4-butoxyphenyl)-3-chloropropan-1-one. The target compound was finally synthesized by reacting this intermediate with piperidine hydrochloride in the presence of triethylamine. The developed synthesis proceeds under mild reaction conditions with simple work-up procedures, affording dyclonine hydrochloride in 60% overall yield (calculated from phenol).
